Formulation and Evaluation of Floating Multiparticulate Drug
Delivery System of Glipizide
Gastroretentive floating multiple-unit drug delivery system for glipizide as a model drug was
developed for prolongation of gastric residence time. Floating microspheres were prepared by
ionotropic gelation method using polymeric material such as chitosan. D-optimal design was
utilized to investigate the joint influence of two variables: drug to polymer ratio (X1) and
concentration of effervescent agent (X2) on the drug entrapment efficiency, percentage buoyancy,
and cumulative percentage drug release. Particle size and surface morphology of prepared
microspheres were characterized by optical and scanning electron microscopy respectively.
Formulated microspheres exhibited prolonged drug release and remained buoyant for more than
